Abstract
Five popular local potato genotypes namely Shil Bilati, Lal Pakri, Shil Pakri, Jam Alu and Pahari Pakri along with one exotic potato genotype Kufri Shinduri were treated with GA3 (400ppm) for collecting maximum number of sprouts in a short period of time during harvesting season of potato with maximum dormant stage. After sprouting, sprouts were used to collect meristems from sprout tip (0.5 mm) to induce callus and regenerate shoot from meristem culture. Meristems were inoculated in six levels of hormonal combinations with GA3 0.5 mg/l, 2,4-D 0.5 and 1.0 mg/l and Kinetin 0.5, 1.0 and 1.5 mg/l along with MS media produced calli. Maximum size of callus (0.82 cm) was observed when meristem of Kufri Shinduri was inoculated in 0.5 mg/l GA3+ 1.0 mg/l 2, 4-D + 1.5 mg/l Kinetin. However, no callus was found to initiate in fresh MS media. The callus texture of Kufri Shinduri and Jam alu was non friable and other four genotypes developed friable calli. Colors of different four calli developed looked to be similar to their mother genotypes. In shoot initiation, Shil Pakri took minimum days (2days) with 0.5mg/l GA3+ 0.5mg/l 2, 4-D+ 0.5 mg/l Kinetin. The maximum shoot length (5.88cm) was recorded in Shil Pakri in 0.5mg/l GA3 + 0.5mg/l 2, 4-D + 1.5 mg/l Kinetin at 30 days after initiation. Thus Kufri Shinduri and Shil Pakri were most potential to generate callus and shoot. Further research should be done for Shil Bilati and Pahari Pakri which showed poor performance during this investigation.
